13 <p>Development of the tomato EST database is
14 currently in progress and continues to be updated on a
15 regular basis as new sequences are added. Our objective
16 is to ultimately make available to the research community
17 a complete set of non-redundant tomato ESTs. The "in
18 progress" collection is continually updated as new EST
19 sequences are added to the database. In some cases new
20 EST sequences provide information that modifies the
21 previous "in-progress" non-redundant EST collection. For
22 example, when clones are found through additional
23 sequencing to be chimeric, or when a new EST sequence
24 "links" two clones previously thought to be unique. In
25 short, the "in-progress" non-redundant set is expected to
26 contain a considerable number of duplications and errors
27 of various types. Once the sequencing phase of this
28 project is completed (fall 2001) a final build will be
29 created and a final non-redundant gene set defined. This
30 collection is currently estimated to range from 25,000 -
31 40,000 cDNA clones which will be condensed, quality
32 confirmed, and re-arrayed into a collection that can be
33 distributed in whole - or can much more efficiently be
34 searched for individual clones than is currently the case
35 with over 25 libraries each containing 4000 - 15,000
38 <p><strong>We realize the need for access by the research
39 community to EST clones prior to completion of this
40 project and thus have instituted a policy whereby up to
41 150 clones per laboratory per year can be requested on a
42 subsidized re-charge basis through the Clemson University
43 Genomics Institute (CUGI) which handles maintenance and
44 distribution of the tomato ESTs and receives funding
45 through this project to offset some of the associated
48 <strong>Cost Basis</strong>
49 <p>CUGI received a total of \$100,000 (apx. \$80K in direct
50 costs) spread over 3 years in support of clone
51 maintenance and distribution. These funds have been used
52 for a) purchase of 3 -80 freezers and service contracts
53 to insure their operation b) support a half-time tech
54 position, c) annual replication of all EST libraries to
55 insure against loss of clone viability, and d) software
56 upgrades used in clone request tracking and eventual
57 public dissemination of all clone requests. It is
58 important to realize that with 22 ordered EST libraries
59 (each containing 4,000 - 15,000 ordered clones) tracking,
60 localizing, and picking each clone represents a
61 significant effort.</p>
63 <strong>Clone Request Charges</strong>
64 <p>Up to 150 clones can be requested per laboratory per year
65 at the subsidized rate of \$5 per clone. An unlimited
66 number of additional clones can be requested by any one
67 laboratory in a 12 month period but at a non-subsidized
68 rate of \$20 per clone. This later price is in line with
69 other clone distribution centers, such as the American
70 Type Culture Collection (ATCC), which recover total costs
71 through re-charge fees. Cost recovery for the entire
72 non-redundant set will be determined when said collection
73 is completed (est. late 2001).</p>
